Yes. It will take some work do so, but it can be done. The code is not
currently very compatible with Win32, but we're working on that for
Bugzilla 2.18. OTOH, if you know your IIS and Windows throughout, setting
up Bugzilla on Win32 isn't that hard. I've done it dozens of times and it
now takes me about 20-30 minutes per installation, but it wasn't all that
fast for the first time ;-)

No. The current bugzilla guide's Windows installation instructions suck.
We have a bug on making them better, though; see the patch on
http://bugzilla.mozilla.org/show_bug.cgi?id=145965. It isn't perfect yet,
but it's a good starting point - especially if you're ready to go with the
Bugzilla 2.16 release candidate 2, for which the instructions are aimed at
(and I highly recommend you to go with the 2.16 series from the
beginning). Also note that securing your Bugzilla installation on IIS
takes some extra measures; with Apache that part is much easier.

The current biggest problem is making the email notifications work. There
is a patch for that and it worked fine the last time I tried, but it may
have bitrotten since. It's attached on
